- Baloxavir is a different mechanism - cap-dependent endonuclease (PA subunit) inhibitor, single oral dose, blocks transcription initiation
- Mechanism dictates the timing: the drug stops new rounds of replication, so it must be given while replication is still happening - within 48 h
