Half-life calculation from clearance and volume of distribution

- t1/2 = 0.693 x Vd / CL -- half-life is determined by clearance and volume of distribution, it is not an independent primary parameter
- Rearranged, this means CL = 0.693 x Vd / t1/2, and Vd = CL x t1/2 / 0.693 -- any one of the three can be derived if the other two are known
- A drug's half-life can be prolonged either by decreased clearance (renal/hepatic impairment) or increased Vd (e.g. oedema, obesity for lipophilic drugs) -- same observed effect, opposite underlying cause
