Definition

  • Polygenic inheritance describes traits/conditions influenced by the combined small effects of many genetic variants, each individually contributing only a modest increment to overall risk - a specific subtype of multifactorial inheritance focused purely on the genetic (not environmental) contribution
  • Increasingly quantified through polygenic risk scores (PRS), which aggregate the effect of many common variants (identified through genome-wide association studies) into a single risk estimate - an emerging tool in several common disease areas, including cardiovascular disease and some cancers
