- Loss-of-function filaggrin gene mutations are the strongest known genetic risk factor for atopic dermatitis -- present in a substantial proportion of moderate-severe cases
- Barrier defect -> increased transepidermal water loss and increased allergen/microbial penetration -> downstream Th2-skewed inflammatory response
